How we started using Bravecto

And so we gathered a dog in Latvia, in a region where there are sooo many ticks. For Hasya it was scary, because ticks for us remained an unresolved problem.And they remembered about Bravecto. Once again we talked with our veterinarian, re-read the studies on the Internet ...

So, the main points that motivated us to give the pill:

  • promised 99% tick protection;
  • according to American studies, none of the tested dogs had any side effects (although it seems that in the European version they write about a possible decrease in appetite, apathy, etc.);
  • principle of action: at the time of a bite, the tick is paralyzed due to the active substance (fluralaner). And we decided. At that time, our dog was 6 months old, he weighed somewhere around 20-20.5, but we gave him a tablet for 10-20 kg (now we take tablets for large dogs weighing 20-40 kg).

Application. The tablet is given during any feeding. If the dog suddenly refused to eat it (and ours has become more choosy with age and does not always eat it voluntarily), you can put it deeper into the mouth and hold it closed until the tablet is swallowed.

Also, Bravecto can be taken with drops / collars / sprays, etc. from ticks, anthelmintic drugs, antibiotics, and almost any other drugs, this does not cause any unwanted reactions and does not reduce the effectiveness of the pill.

The next few days after the first application, I looked at the dog more than ever, but there were no changes in behavior, in activity, in appetite, or in stool. A dog is like a dog.

A few days later we went to the lake in the forest 🌲🌳🌲, where, as it turned out later, there were darkness with ticks (we even bit a person). I have a German shepherd, a large male lohmach weighing about 50 kg., Age 4 and a half years. In recent years, they used Rexolin Plus drops, but last year interruptions began in Ukraine with them, the manufacturer changed, after that there were negative reviews about the drug and this year the question for me was how to protect the dog from ticks further (first of all) .

For the first time about a miracle tablet, “Bravecto” I heard from a salesman in a pet store, as a novelty in the Ukrainian market and as a drug that has proven itself well abroad. Using the Internet, I found out for myself that the tablet manufacturer is a well-known Austrian company MSD, which widely advertises its products.

It first appeared on the market of the CIS countries in Belarus, Russia and Kazakhstan, where it was officially approved for use, and from this year it appeared in Ukraine. I bought a tablet in the ZOO online pet store ([link]), and they were from the official distributor in Kiev - “VETBIO” so that the tablet was real (checked). When I offered the dog a pill, he didn’t want to eat it; I had to put it back on the root of the tongue feed him his head.

And after 6-8 hours after a walk in the wasteland, when he was going to take Sobakevich in the evening for an evening walk, he noticed a tick biting his head on a dog’s head near his ear. Thus, as a result of using the miracle of the Bravecto tablet, I came to the following conclusions that at the first contact with ticks on the 18th day of using the tablet, the tick digged into the dog and infected it and remained alive. So I really regret that I was seduced by the laudatory reviews about the pill, that it was supposedly “not punched” by ticks.

Nonsense! I was convinced from my experience. In addition, the same was confirmed in the veterinary center, saying that they are already their second such "infected" under the tablet "Bravecto". And it turns out that the tick introduces babesia when the dog bites along with the saliva where they are.

Thank you for your health, but I bought a tablet (expensive) in order to preserve this health of the dog, and it turns out that it was lost due to unscrupulous advertising by the manufacturer. The Bravecto® tablet form is a new way to protect dogs of all breeds from blood-sucking parasites. The preparation contains active substances that protect the four-legged pet from ectoparasites and dangerous diseases transmitted by fleas, mosquitoes, ticks for 12 weeks.

Operating principle

An antiparasitic agent with the trade name Bravecto® (Bravecto) is a tablet form for oral administration. A new drug in the veterinary market is classified as a systemic insecticide and acaricide.

Pharmacodynamics

Absorption of an active substance by the digestive tract is very high. Fluralaner penetrates the systemic circulation and reaches a critical mass in blood plasma during the day with bioavailability of up to 20-27%. Feeding a pet before giving a pill enhances the absorption of the components of the drug.

Insectoacaricidal concentration of the active substance in the blood plasma lasts up to 85 days. All this time, it has a detrimental effect on ectoparasites. However, in the event of a decrease in immunity, demodicosis can develop into an unpleasant disease, which is manifested by spontaneous alopecia of a localized or generalized nature.

Demodecosis complicated by a secondary microbial infection can cause serious lesions in the form of severe scratching and purulent inflammation of the skin. An advanced treatment for generalized dog demodicosis is the use of Bravecto chewable tablets.

At the same time, to cure this difficult disease, it is enough to take one Bravecto tablet.But if the manifestations of a bacterial infection are also pronounced, in addition to Bravecto, it is also advisable to use antiseptic shampoos and sometimes antibiotics.

The tick that lives in the ears of our four-legged pets is called otodectes, and the disease that it causes is called otodectosis. Puppies, especially small breeds, are subject to otodectosis, according to veterinarians, adult dogs are less likely to suffer from otodectosis. Infection with otodectosis occurs as a result of contact with a sick individual.

Before starting treatment, the patient’s ears must be thoroughly rinsed and rid of the accumulated sulfur with special hygienic solutions and drops. If bacterial inflammation of the skin has already developed on the background of otodectosis, then the use of antibacterial ear drops is necessary.

The next step in the treatment of otodectosis is the use of anti-tick drugs, and Bravecto is not suitable for this case. Ear drops against parasites are quite common, however, are an ineffective treatment and cause great discomfort when instilled into the ears.

Drops at the withers, such as the Inspector or Lawyer Stronghold, are a more effective way to combat otodectosis. To completely cure otodectosis, with the exception of possible relapses, Inspector drops are used twice with an interval of 1 month. A more reasonable approach is the use of repellent drugs or agents that can quickly kill ticks before transmission of pathogens, preferably with a long duration of action.

Fluralaner (Bravecto ™) - an innovative ectoparasiticide belonging to a new class of compounds, isoxazolines, is effective against Ixodes ricinus, Ixodes scapularis, Dermacentor reticulatus, Dermacentor variabilis and Rhipicephalus sanguineus, i.e. against all types of ticks capable of transmitting the most important pathogens of human and domestic animals, in particular anaplasmosis, babesiosis, borreliosis and ehrlichiosis.

The mechanism of action of fluralaner is based on the blockade of ligand-dependent chloride channels (both GABA receptors and glutamate receptors), which leads to suppression of the arthropod nervous system, which leads to paralysis and death of fleas and ticks.

Fluralaner has high selectivity for arthropod neurons compared to mammalian neurons and is well tolerated by dogs from 8 weeks of age, including dogs with a mutation in the MDR-1 (- / -) gene.

In dogs, fluralaner is characterized by a long half-life, a long retention time, a relatively high volume of distribution and low clearance. The long retention time of fluralaner in the dog’s blood plasma causes stable lethal activity against fleas and ticks for 12 weeks after a single oral administration.

For the action of fluralaner, it is necessary that the ticks attach to the skin of the host and, thus, swallow the active component. Continuous feeding increases the risk of transmission of pathogens from ticks to dogs, however, infection usually does not occur immediately after the tick is attached to the dog.

For the transmission of most diseases transmitted by ticks, initial attachment and a period of bloodsucking of at least 24-48 hours, during which pathogens reactivate, are necessary. This period between tick attachment and transmission of the pathogen allows fluralaner to exert its effect.

If infected ticks die during this period, then the transmission of pathogens is most likely to be prevented. Thus, the rate of extermination, defined as the time required to kill already attached ticks or to kill ticks after repeated invasion, is an important factor in the prevention of tick-borne diseases.

Based on the justification indicated above, 2 studies were conducted to assess the rate of tick destruction by fluralaner. For this, the efficiency of killing ticks was measured after 4 and 8 hours (study 1) or after 12 and 24 hours (study 2) after a single use, as well as at various intervals after re-infection of these dogs during the 12 weeks of the drug.

In both studies, I. ricinus ticks were selected to infect dogs.

Study design

Studies have examined the rate of lethal action of fluralaner on I. ricinus ticks in dogs. The study included a total of 48 healthy adult Beagle dogs (<7 years old) who were not treated with any antiparasitic drug for at least 12 weeks before the start of the study.

Before distribution into groups of dogs weighed (range 9.3-19.9 kg) and conducted a clinical examination. Participating dogs had previously been infected with 80 fleas (Ctenocephalides felis) to demonstrate their susceptibility to parasitic infestation and to confirm the absence of ectoparasiticides.

Dogs were ranked in decreasing order of fleas (in the range of 63-80 fleas / dog) and randomly assigned to 8 groups (4 treatment groups and 4 corresponding control groups) of 6 dogs using a computer-generated randomization list.

All dogs were kept indoors. During periods without parasitic infestation, dogs were kept in the groups in which they were distributed, and during periods of parasitic infestation, all dogs were kept separately.

The temperature in the vivarium with dogs was 17-22 ° C, and relative humidity - 40-90%.Dogs were fed standard, commercially available dry dog ​​food once a day, and access to water was not restricted. General health observations were made once a day throughout the study.

Drug use

On day 0 (i.e., the day the drug was used), dogs in 4 experimental groups received chewable tablets of fluralaner based on the body weight of a particular animal and a minimum dose of 25 mg fluralaner / kg of body weight.

Fluralaner chewable tablets were placed in the back of the mouth on the tongue to stimulate swallowing. Dogs received half the daily diet no earlier than 20 minutes before using the drug, and the rest immediately after.

Each dog was continuously monitored for 1 hour after administration of the drug to determine whether the chewable tablet would spit out or burp; such cases did not occur. Dogs in 4 control groups were left without treatment.

Tick ​​infection and its assessment

The ticks I. ricinus used in the study were grown in the laboratory for at least 2 generations after selection from natural habitats in Europe. Dogs under the influence of the sedative drug medetomidine were infected with ticks at 2, 28 (4 weeks), 56 (8 weeks) and 84 (12 weeks) days.

At each infection, 50 non-fed adult female ticks were placed directly on the hair of each dog along the back, on the side and on the head. In addition, 10 Ixodes ricinus males were placed on the dogs to provide the females with optimal conditions for attachment. Tick-infected dogs were kept in individual pens until parasites were removed.

Tick ​​load (the number of males and female ticks) in all dogs in each pair of the experimental and control groups was evaluated through 4 (± 0.75), 8 (± 0.75), 12 (± 1.5) or 24 (± 1, 5) hours after treatment (week 0) or after re-infection (on the 4th, 8th and 12th week).

The entire body of each dog was inspected, and ticks were carefully removed with forceps. Ticks were classified as dead or alive, attached or unattached, and counted. The personnel who classified and counted the ticks were not aware of the treatment prescribed for each dog.

Statistical analysis

Statistical analysis was performed using the SAS® software package (SAS Institute Inc., Carey, North Carolina, USA, version 9.2). In all statistical calculations, the statistical unit was a single animal. Percentage efficacy against ticks was calculated for each treatment group and each evaluation time in accordance with the Abbott formula:

The significance of differences was evaluated by comparing the logarithm of the number of live ticks in each treated group with the logarithm of the number of ticks in the corresponding control group for each time of assessment.

Treatment groups and control groups were compared using a mixed linear model, in which the group of animals had a fixed effect and the block a random effect. The two-sided confidence level for the F-criteria of the model effects was chosen equal to a = 0.05.

results

In none of the 24 dogs treated with fluralaner, treatment-related adverse events were found during the 12-week period after treatment. The effectiveness of the destruction of ticks was 89.6% after 4 hours, 97.9% after 8 hours and 100% after 12 and 24 hours after treatment.

Eight hours after reinfection, the efficacy was 96.8%, 83.5%, and 45.8% 4, 8, and 12 weeks after infection, respectively. After 12 and 24 hours after repeated infections throughout the entire 12-week study period, the efficiency of tick kill was at least 98.1%.

In dogs receiving fluralaner, the number of ticks at 4, 8, 12, or 24 hours after treatment was significantly lower (p <0.0001) than in control dogs not receiving treatment. In dogs treated with fluralaner, the number of ticks at 8, 12 and 24 hours after re-infection was also significantly lower (at all time points p <0.0001; 8 hours after re-infection at week 12, p <0.004) than in control dogs. 4 hours after re-infection at week 4, a significantly lower number of ticks was also recorded (p <0.03).

The discussion of the results

Fluralaner (Bravecto ™) is the first ectoparasiticide for oral use, showing a significant duration of action against ticks. This study clearly shows that fluralaner quickly destroys ticks with increasing efficiency over time, and the full effect is achieved after 12 hours and lasts for the entire 12-week period of action.

Fluralaner is a systemic parasiticide and acts on the tick with the beginning of feeding on the host body. A quick lethal effect limits the feeding period. Usually, transmission of pathogens to the host does not occur immediately after tick attachment, but after prolonged bloodsucking.

Before transmission, pathogens are activated directly in the salivary glands, or they need a reactivation period for reproduction and migration to the salivary glands. This reactivation period begins after attachment to the host, and transmission occurs when the tick burps excess fluid into the wound from the bite.

Several studies have been conducted to evaluate the time required for transmission of the pathogen. Infected ticks were allowed to feed on the host's body for certain periods of time.

After removing ticks, the host was checked for the presence or absence of a pathogen or specific immune response. If the data indicated that the host was in contact with the pathogen, then the time spent by the tick on the host was considered sufficient to transmit the pathogen.

Thus, transmission of B. burgdorferi to mice by ixodid tick nymphs occurred no earlier than 24 hours after attachment: 0 out of 18 mice were infected after contact with B. rurginus nymphs infected with B. burgdorferi for 24 or 48 hours, and not one of 58 mice did not become infected after exposure to B. scurgularis nymphs infected with B. burgdorferi for 24 hours.

Transmission to dogs of Babesia canis from infected D. reticulatus ticks occurs no earlier than 48 hours later. Moreover, Prullage et al. concluded that the transmission frequency of B. burgdorferi, A. phagocytophilum and B. microti is significantly reduced if ticks are not allowed to feed for more than 24 hours.

After the transmission becomes possible, the probability of transmission of the pathogen increases significantly and continues to increase with increasing duration of bloodsucking. When comparing data on the time of transmission of pathogens with the results of our study, we can conclude that fluralaner kills ticks before transmission of pathogenic borrelia and babesia begins.

Pathogens such as A. phagocytophilum and E. canis accumulate in the stomach, but they can also be found in the salivary glands, which makes it possible to transmit earlier. Thus, transmission of A. phagocytophilum to mice from nymphs I. scapularis occurs during the first 24 hours, and transmission of E. canis occurs within the first 3-6 hours.

Thus, the risk of transmission of these pathogens cannot be completely excluded taking into account the rate of destruction of ticks by fluralaner and published values ​​of transmission time, according to laboratory studies.

However, these studies were conducted under laboratory conditions; It is not possible to reproduce all available variables of the natural state of the carrier and the host (e.g., invasive pressure from the carrier and the prevalence of infection among carriers, interruption of tick feeding, differences in the age, breed and immune status of the host, as well as the presence of other reservoir hosts) in the laboratory.

More importantly, in some studies on dogs, the use of commercially available acaricidal drugs protected against infection with A. phagocytophilum or E. canis.

According to the description, these drugs have a lower kill rate (i.e., the time required to kill> 90% of attached ticks after treatment / reinfection is longer) than fluralaner. Thus, fluralaner appears to have no less ability to protect dogs from infection with A. phagocytophilum and E. canis.

The systemic mechanism of action of fluralaner provides additional benefits for the user compared with substances intended for local application or used as part of collars and remaining on the surface of the dog’s body.

When using these drugs, a decrease in effectiveness against ticks due to loss of a collar or after bathing is possible. In addition, the fact that ticks die after feeding on a dog receiving fluralaner also reduces the likelihood of infection of another dog, which can occur when using substances with an exceptionally repellent effect.

In addition to the high rate of destruction of parasites and the systemic mechanism of action, the long duration of action of fluralaner brings additional benefits to owners, since it allows less frequent use of the drug compared to other commercially available acaricides for monthly use.

Frequent and regular use of ectoparasiticides is an important tool for the prevention of tick-borne diseases. However, the owners do not observe the regimen well enough, which leads to periods of lack of protection of dogs from ectoparasites, i.e. cases of non-use of funds for monthly use with the recommended frequency.

This puts at risk not only the health of the dog, but also other dogs and people, since an unprotected dog is a potential reservoir of pathogens. Therefore, an ectoparasiticide with a high rate of destruction of parasites and a long duration of action - fluralaner (Bravecto ™) - helps to prevent and control diseases transmitted by ticks.

After application, fluralaner destroys ticks quickly - after 4 hours, and throughout the entire 12-week period of action, it provides an almost complete lethal effect against ticks within 12 hours after infection.

During this spring-summer period, she did not remove live ticks from dogs. I found on a small (he has short hair) two dried ticks, that is, they bit him, but then they died.
